Abstract 摘要

胎牛血清(Fetal Bovine Serum, FBS)廣泛應用於細胞培養中,作為提供營養、增長因子、激素、結合/運輸蛋白以及細胞貼附因子的重要補充物。然而,在先進療法醫藥產品(ATMP)的生產中,減少或替代 FBS 是當前的重要挑戰之一,尤其是在 Vero 細胞的培養與生產上。

本研究探討了 蛋白水解物(Peptones) 是否能夠作為 FBS 的替代物,用於 Vero 細胞的擴增培養基。研究結果顯示,蛋白水解物的原料來源與製程 會顯著影響 Vero 細胞的增殖能力。

血清減量 50%:透過補充 0.5 g/L 蛋白水解物可將血清濃度從 10% 降至 5%,且仍能維持相同的細胞增殖效率
蛋白水解物的來源影響表現:來自酪蛋白(Casein)、小麥(Wheat)和大豆(Soy)的蛋白水解物對 Vero 細胞的影響各不相同。

製程影響蛋白水解物的效能

  • 伽馬射線(Gamma Irradiation)與超濾(Ultrafiltration) 可提升小麥蛋白水解物的效果。
  • 酪蛋白水解物 則在相同處理條件下效果較差。

最佳化培養基搭配蛋白水解物可獲得最佳效果,比在一般培養基中單獨添加蛋白水解物表現更佳。

Investigating the Proliferative and Serum-Reducing Effects of Various Protein Hydrolysates on Vero Cells for ATMP Bioproduction

Fetal bovine serum is widely used in cell culture as an additive to media. Serum is a provider of different elements such as nutrients, growth factors, hormones, binding/transport proteins, or even attachment and spreading factors.

In the present work, we investigated whether peptones can be used as an alternative to fetal bovine serum in an expansion media for VERO. Data indicates that the raw material and the process used to manufacture peptones have an impact on the proliferation of VERO.

Moreover, whether it will be combined with casein, wheat, or soy-based peptones, we can reduce the serum by 50% and still obtain the same performance regarding the proliferation using 5% of serum plus 0.5 g/L of peptones.

The best peptones were combined and tested with different media: DMEM 10% FBS (control media) versus a novel optimized Vero media: BMBD145 Av.

These results illustrate how satisfactory is the batch-to-batch consistency of casein-based peptones. Furthermore, one batch of wheat-based peptone was given different post-process treatments. It appeared that gamma irradiation and ultrafiltration have a positive impact on proliferation. For casein-based peptones, it turned out to be the opposite.

Plus, peptones give a better result when used with an optimized media rather than with a general one.
